Russell was born to Stephanie (Stephens) and David Harold Russell, an executive at Nissan Motors. She began her career on the Mickey Mouse Club in 1991. She stayed with the show until 1993. After leaving The All New Mickey Mouse Club (where she, along with her casemates recorded an album, MMC), she moved to LA to pursue an acting career. She starred in Aaron Spelling's Malibu Shores (1996) on NBC.

Russell's breakthrough role came in 1998 where she played "Felicity Porter" in the WB's popular series Felicity (1998-2002). In 1999, Keri won her first Golden Globe award for her role in Felicity (1998) in which she continued until the show ended in 2002. Keri pursued a movie career starring alongside Mel Gibson in We Were Soldiers (2002) and Joan Allen in The Upside of Anger (2005). In 2004, Keri made her Broadway debut in "Fat Pig" alongside Andrew McCarthy and Jeremy Piven.

In 2006, Russell became engaged to Shane Deary, a Brooklyn-based contractor she met through mutual friends.[11] They married in New York on February 14, 2007.[28] They have two children: a son born in 2007 and a daughter born in 2011.[29][30] Russell and Deary separated in early 2013 and divorced in mid-2014.[31][32] Since 2014, she has been in a relationship with Welsh actor Matthew Rhys, her co-star in The Americans.[33] They have a son, born in 2016.[34][35] In interviews conducted in 2021, Russell and Rhys referred to each other as husband and wife,[36][37] although in an interview in 2025, Rhys said "we literally haven't got round to marriage yet".[38]
