Duplication

This page has a lot of duplication from the CRISPR page (therefore delete from here), and some material that would be better to mainly be on the CRISPR page e.g. transcription. I intend to start this soon.

Misc

Cas9 should probably have its own page, given how many papers are being published using the protein outside of its original context in the CRISPR system. Any objections?

Cleavage

This is recent science, and apparently written by those in the know, for those nearly in the know. At any rate it uses WP:JARGON, such as "cleave" not defined or explored in linked explanatory articles. If those more basic articles do not yet exist and will not soon be written, then this and other terms ought at least be briefly defined at an early appearance in this article. Occurence of Cas9

I have a source listing at least 3 other bacteria using Cas9 (https://www.nature.com/articles/s41579-019-0299-x/figures/2 from https://www.nature.com/articles/s41579-019-0299-x), but since only so many bacteria have been sequenced yet, there are likely many more. Too focused on Cas9 from Streptococcus pyogenes so pls add some orthologs

I don't have time to overhaul this (editing some other pages rn), but just wanted to let everyone know that at the time of posting this, the page is wayyyyy too centered on Streptococcus pyogenes's Cas9 (SpCas9).

The protein has a bunch of orthologs, some of which have different PAMs or even different cleavage properties. For example, the one from Francisella novicida (FnCas9) makes sticky rather than blunt ends. These have been used in a variety of genetic modification studies. They've even been engineered for a more desirable function (converted into a nickase, less of a stickler about the PAM, probably some other stuff I'm too lazy to include).

This page seems to get a lot of Wiki Education edits, so unless someone beats you to it, got some free suggestions for you. RenaMoonn (talk) 03:01, 11 June 2026 (UTC)[reply]
